ATC-vet classification code: QN05AA04 Acepromazine.

Acepromazine maleate is a neuroleptic of phenothiazine group, sedative drugs. Sedation consists in reduction of sensitivity to external factors. The action is aimed at blocking dopaminergic receptors of different levels in the central nervous system.

Its application in the form of 1% solution for injection provides a calming and myorelaxing effect by reducing the excitation of the central nervous system.
At the same time the drug has hypothermic, hypotensive, antihistamine, adrenolytic, antispasmodic, antiemetic action, accelerates and enhances the effect of sleeping pills and local anesthetics.
When administered intravenously, the action of the drug is seen after 5 to 8 minutes, and when administered intramuscularly – after 15 to 25 minutes. The sedative effect lasts for up to one hour.
When parenteral administration acepromazine maleate quickly penetrates into all tissues.

The active substance of the drug belongs to lipophilic compounds, has high volume of distribution and high degree of binding to blood plasma proteins (up to 90%), especially to albumins. Once in the body, acepromazine is well distributed in all organs and tissues.

Acepromazine maleate is metabolized in the liver and excreted in the urine, mainly as a glucuronic acid metabolite.

Application :

The drug is administered to animals for premedication before surgical intervention 15-20 minutes before the main anesthetic, pain relieving, as sedative for aggressive animals before examination and transportation, spasmolytic – for horses (in case of spastic colic).

Method of administration and dosage

The drug is used once, injected slowly intravenously or deeply intramuscularly, for stallions and cats only intramuscularly.

Doses of the drug for various animal species are shown in the table.

Animal species Method of administration Dose, ml / 22lb  (10 kg) of body weight
Horses slowly intravenously 0.05-0.06
deep into intramuscular injection 0.05-0.1
Dogs slowly intravenously 0.05-0.1
deep into intramuscular injection 0.2-1
Cats deep into intramuscular injection 0.5-1

Contraindications :

Hypersensitivity of the animal to the drug.

Do not use in animals with liver, heart, lung function abnormalities, diabetes, and pregnant females!

Do not use together with organophosphates (insecticides)!

Do not use together with local anesthetics!

Do not use in case of hypothermia, hypotension, bleeding with significant blood loss!

Do not use in intoxication with strychnine!

Overdose (symptoms, emergency measures, antidotes)
Avoid overdose!

Symptoms of hypotension occur in case of overdose.

In case of overdose symptomatic measures are taken.

As antidotes, the use of a-adrenomimetics and respiratory analeptics is recommended.

Caution :

Slowly administered intravenously!

Rapid administration can lead to cardiac arrhythmia. Stallions are advised to use the drug only intramuscularly, observing the minimum dose, due to possible penile prolapse, which usually lasts up to 2 hours and does not require treatment. If penile prolapse lasts more than 24 hours, the animal needs the help of a veterinarian.

The drug has no analgesic effect at all, an additional analgesic should be used for pain relief.

It is necessary to take into account the increased sensitivity to the drug of some breeds of dogs, namely: greyhounds, dogs of large breeds, boxers and other representatives of brachycephalic breeds. Increasing the dose does not lead to prolongation.

Side effects :

Possible phenomena of bradycardia, constriction of airways, slight decrease in body temperature. During rapid administration, cardiac arrhythmia may occur.

Priapism and penile prolapse in stallions (sometimes leading to permanent paralysis).
